micro-spectroscopy of collagen-containing tissues: the impact of optical

Conference RoomRaman spectroscopy as one of the optical spectroscopic methods
provides valuable information about the molecular structure and conformation,
which gains great attention for diagnosis and monitoring of biological
objects. All biomedical optics including Raman spectroscopy are suffering
from strong light scattering or limited in-depth light penetration. These
issues pose significant hindrance to the practical application of the optical
techniques in clinical practice. In the 90s of the last century, the optical
clearing technique based on the immersion of the tissue into optical clearing
agent was introduced and developed to enhance the detection depth using
optical techniques. In this thesis work, I studied the impact of optical
clearing method on collagen-related Raman peaks to control the optical
properties of biological objects, specifically ex vivo porcine dura mater and
skin-dermis in-depth using confocal Raman micro-spectroscopy. I determined
the impact of pure glycerol as an optical clearing agent on the intensity of
collagen-related Raman peak, as well as on dehydration and dissociation of
the collagen using confocal Raman micro-spectroscopy at various depths
ranging from 0 to 250 μm and treatment times [1]. In addition, the
diffusivity of 50% glycerol and the changes in the water content within the
collagen of the dura mater were monitored at various depths and treatment
times [2]. The effect of the optical clearing on the Raman spectra recorded
with 633- and 785-nm excitations in ex vivo porcine skin-dermis was also
studied at different depths and treatment times, together with the influence
of dimethyl sulfoxide as an enhancer additive [3].[1] Ali Jaafar, Maxim E.
